You need to add python to your Build-Depends line in debian/control.

I suggest using sbuild or pbuilder to test your build in a clean chroot.
You can't effectively test whether your Build-Depends are correct by
just running dpkg-buildpackage in your normal system, because any
development system is going to have far more than the basic
build-essential set of packages already installed.
