I just made a fresh install according to
http://www.sikuli.org/download.html on my Win 7-32 even without
rebooting and everything worked as expected.

So you might try, to get rid of everything (including the registry entry
according to faq 2005), reboot the system and start all over again.

If you have done everything according to faq 2005, then I have no other
idea.
