My advices:

1. Use the build in Sikuli-IDE !

2. ignore regions for now
2.1 if you are having more than one screen: move the game-window to the screen Sikuli uses as primary (you got the right one, if your script finds the element you are looking for at runtime)

4. Troubleshooting #2: search http://doc.sikuli.org

5. Troubleshooting #3: ask your question here

If you don't wont to do any real fancy stuff you probably don't need any
Python knowledge
