I would strongly advise against going down that path, while its certainly possible to remaster an ISO by replacing files within the sqaushfs, it
is a terrible idea and is only going to cause problems for your users (and probably false bug reports for our project).
The right way to do this, is to make customisations within the relevant debian packages (see for example ubuntu-gnome-default-settings package),
host these on a ppa, then use live-build to generate the new ISO's. You would also need to fork the ubuntu-gnome seeds and ubuntu-gnome-meta
package also.
