Sickly does not know that it is a game, it is just pixels in rectangular areas and images.
So wether a script is able to automate something reliably depends on the creator of the script and the knowledge he has about how Sikuli works and what the useable features are.
Sickly can just automate what you see on the screen.

... and yes you can restrict the search region (I already mentioned

This a very vague description of what is happening.
I mentioned some aspects in comment #1, that might help to debug the problem.

BTW: In games one often has buttons that periodically change their
outfit (e.g. glow), if this is the case here, you have a problem, that
needs an intelligent approach. did you already scan the net (google)
about "Sikuli and gambits"? might be worth to do that.

One more thing: your script is a simple row of clicks and waits, but at
no point I can see that you check, wether the game reacts in the way you
expect it.
